Some glyphs have conditions to satisfy, like having a minimum of 25 Dexterity in a particular area on the Paragon Board. Certain glyphs also require a certain number of Primary Stat nodes to be activated, which can be difficult to attain. Certain glyphs also have a radius, which increases as you level it up.

Glyphs can be applied to many different items and weapons, except for those of a specific class. Some of these glyphs have additional requirements, like a certain amount of Dexterity damage, or Attack damage on a specific portion of the Paragon board.
